Collagen

1) Most abundant protein in the body

2) Major structural protein in ECM

  • Tropocollagen is the monomer unit
  • Consist of 3 α chains – left hand structures but on combination right handed triple helix

Components:

Glycine residue, -OH proline & OH-lysine

Gly-x-y repeat

Synthesis:

Endoplasmic recticulum of fibroblast

Stage 1:

Formation of α-chains → Combine → Pre-pro-collagen

Stage 2:

Hydroxylation of Proline & Lusine

Glycosylation to OH- Lysine

Intracellular event- convert → Pre-pro-collagen → Procollagen

Extracellular event

Stage: Tropocollagen is formed

Types:

Type 1: Skin & bONE

Type 2: CarTWOlage

Type 3: ArTHREE

Type 4: Basemnet membrane floor

Disorders of Collagen:

Type 1:

  • Osteogenesis Imperfecta aka Brittle bone d’s
